Angiotensin-II and angiotensin-III in the mechanisms of alcohol motivation in rats: comparative aspects

A-II and A-III are the major effectors peptide of the renin-angiotensin system with similar physiological activity (initiation of thirst, hypertension, regulation of water and salt balance, and others.). Comparative analysis of angiotensin-II (A-II) and product of its enzymatic degradation – angiotensin-III (A-III) involvement in the mechanisms of development and realization of alcohol motivation in rats was provided. For this purpose conjugates A-II and A-III with BSA were used. The effects of active immunization of animals by conjugates A-II and A-III to BSA on the formation alcohol motivation were opposite: in the case of A-II was observed activation, and when using A-III – suppression of its. In turn, this experimental procedure is the same effect on realization of alcohol motivation in animals, and A-II and A-III significantly increases voluntary alcohol consumption. During the development and in the conditions of realization alcohol motivation immunization by A-II decreased the negative consequences of chronic alcohol consumption and A-III – intensified them. Proposed by the authors the hypothesis of an autoimmune nature development of alcohol dependence and mechanisms of selective participation of protein complexes A-II and A-III in this process discussed.
