Effectiveness of the new cortexolone derivatives as anti-craving medications in the rat models of experimental remission from alcohol and drug dependence

To compare the effects of three cortexolone (substance S) derivatives we used an animal model of alcoholism – long-term alcohol consumption followed by deprivation phase in rats. We found that repeated treatment with S5 (cortexolone-acetate-pivaloat) significantly suppressed relapse-like alcohol drinking determined in a "free-choice" paradigm after extended period of abstinence. Furthermore S5 decreased morphine – and cocaine-craving and prevented relapse after long-term period of withdrawal in the "place-preference" model of drug abuse in rats. Possible mechanisms that underlie cortexolone derivatives effects and potential perspectives for S5 to be used in the post-abstinent period as a novel therapeutic intervention for treatment and prophylaxis of the relapse in alcohol and drug-addicts are discussed.
