Introduction. Oxidative stress (OS) is an important link in the pathogenesis of many diseases, including alcohol dependence syndrome. Comparative study of peripheral markers of OS and programmed death (apoptosis) of blood cells in different age groups of alcoholic patients and healthy individuals is important for obtaining new knowledge about the laws of OS development and pathogenetic mechanisms of alcohol dependence.
The aim of this work was to compare the peripheral markers of oxidative stress – oxidized proteins (protein carbonyls, CP), products of lipid peroxidation (TBA-reactive substances, TBA-RS), as well as the indicator of mononuclear apoptosis in different age groups of healthy individuals and alcoholics.
Methods. The material for the study were bloods of 31 alcoholic patients (F10.302) and 31 healthy individuals (all males). The study participants were divided into groups depending on their age: group 1 (under 45 years old) included 13 healthy men and 13 alcoholics, group 2 (over 45 years old) included 18 healthy men and 18 alcoholics. CP and TBA-RS of blood plasma were determined spectrophotometrically. Determination of cell death (mononuclears) was performed by flow cytofluorimetry.
Results. An increase of CP in patients’ blood plasma in comparison with healthy controls’ plasma was found in the younger group (under 45 years): CP in healthy individuals was 0.18 (0.11–0.21), in patients – 0.31(0.22–0.33), *p=0.002. In the group of participants over 45 years of age, there were no significant differences between patients and healthy individuals in both the level of CP and the level of TBA-RS. No differences were found in the apoptosis of mononuclear cells between alcoholics and healthy individuals in both groups: under 45 years and over 45 years.
Conclusion. In alcoholics, the increase in the level of the peripheral marker of oxidative stress – plasma carbonylated proteins – occurs at a younger age compared to healthy individuals, which indicates an earlier formation of oxidative stress in alcohol dependence. At the same time, the indicators of mononuclear apoptosis in patients may remain within the normal range.
