Universal solidphase extraction technology of medicinal and narcotic substances from the hydrolysate of urine for the purposes of gas chromatography – mass spectrometry analysis

Objective: to study the isolation of narcotic and psychotropic drugs and other toxic (medicinal) substances of a weakly acidic, neutral and basic nature from hydrochloric acid hydrolysate of urine on sorbents of the mixed Bond Elut Certify type, in which silanol groups are associated partly with alkyl hydrocarbon radicals and partly with cation exchange substituents. Methods. The effect of the sorbent washing components used in the solid-phase extraction process, the composition of the eluents on the chromatographic background and the reliability of the identification of substances of a weakly acidic, neutral and weakly basic nature was studied by gas chromatography and mass spectrometry. Results. It was found that when the sorbent of phosphate buffer, acetic acid solution, and hexane are excluded from the washings, the gas chromatographic background practically does not change, and only 4 ml of distilled water does not worsen when the mud is washed. The effective desorbing effect of pure Acetonitrile, Methanol, Acetone, and Ethanol on the elution of weakly acidic and neutral substances was determined, and in a mixture of Acetonitrile-Ammonia (9:2), Methanol-Ammonia (9:2) and Acetone-Ammonia (9:2) – substances of a weakly basic nature,Comparable results were obtained both for the desorbing activity of eluents and for the gas chromatographic background. Conclusion. A universal technology of solid-phase extraction of narcotic and psychotropic drugs, other toxic substances, including drugs, from urine hydrolysate on mixed-type sorbents Bond Elut Certify for GC purposes is proposed/MS analysis.
